Hey All
Im currently using 6M HCl to hydrolyse my Protein (6h at 105°C). After that I need to evaporate the HCl, followed by derivatization and GC-MS/MS.
However, if I leave the HCl and dry at 85°C with open caps, I get a brownish/black Pellet and consequently my solution of MSTFA and Acetonitril is colored.
Is there a way to avoid this?
I was thinking by neutralizing. Then again you have salt. Is that problematic for derivatization or GC? Could I use PCR clean up tubes to make it salt free?
Or evaporate under Nitrogen atmosphere?
Or should I just dont care about the color and change the GC inlet more frequently?
