Hi, We are working on 15N2 method to determine Nitrogen fixation by free living in Soil. However, the difference in between samples of natural abundance (NA) and Enriched (EN) by 15N2 is very very close some times NA is higher than EN.

This is quite scary!----15N2 gas from Sigma-Adlrich to measure nitrogen fixation and 15N ammonia/nitrate/nitrite contamination might be the cause.

Maybe the enrichment was too small. I am sure you have seen and followed all recommendations in "Measuring plant-associated nitrogen
fixation in agricultural systems" by Murray Unkovich, David Herridge, Mark Peoples, Georg Cadisch, Bob Boddey, Ken Giller, Bruno Alves and Phillip Chalk. It is available as pdf.
Yes, i have gone through these chapters..by the way I used 20mL vials, placed 5g wet soil in to and flushed by nitrogen for one minute and replaced 10% (~1.5mL) air by 15N2. Incubated for 2 days, 4 days and 8days.
