To select the positive plants, I used 100mg/L glufosinate ammonium solution to spray Arabidopsis seeds each three days. My plants are T2 generation.
The picture attached is one-month seedling.
WTs seem better than mutants. I was totally comfused.
I used glufosinate ammonium from sigama, product number is 45520 (https://www.sigmaaldrich.com/catalog/product/sial/45520?lang=en®ion=NL).
Could anyone know why this is happend? WTs seem not die at all.
The WT plants seem WORSE imo – they have smaller leaves and are flowering, which is a response to stress. The lower plants seem more content lol. Plants often flower in anticipation of death – this is how annual plants function :)

Using basta for in vivo experiments may need higher dosages. If you have enough plants for experiments, try up to 400mg/L. In our experiments, in vivo trials were successful at 300mg/L.
Basta efficiency for in vivo trials depends on different environmental factors (such as heat, moisture, irrigation).
Try this way;
-irrigate soil with basta (100mg/L) 2 days before inseminating.
-use basta again when seedling start to grow.
For fully grown plants, 100 mg/L might be just itching the tissue J You should try higher dosages.
Suggest you do your screening for basat resistance at a earlier stage. We can successfully select basts resistant plants by spraying 150 mg/ lt on 15 day old seedlings . Spray it twice or thrice with 2-3 day gap. Score for phenotype after 7 to 8 days. Susceptible seedlings turn yellow and die . Resistant seedlings stay green and produce new leaves.
