Within ImageJ, how does one find accurate parameters that do not need to be changed while measuring cell migration in a scratch assay?

While analyzing cell migration in scratch assays on imageJ, there has been many inconsistencies in results. Using the same parameters on the same well, imageJ seems to be very inconsistent.
