Will it be too artificial to measure T cell activity in tumor through stimulating T cell with PMA in vitro?

As we all know, stimulating T cell with PMA in vitro can effectively activate T cell unspecificly. If we want to measure T cell activity in tumor, will stimulating in vitro make it too artificial?
