Can anyone help with a problem in detecting over-expressed Flag tagged protein?

I am looking for 25kda flag tagged over expressed protein in western blot. It is a membrane protein. I tried lysis of cells using RIPA and NP40. I can detect the flag with mouse anti flag antibody...
