What should be the treatment duration of ZnSo4 for cell line study?

We would like to replete cells with ZnSO4 to see the effect on expression levels of Zn-transporters. So What can be best time duration for ZnSO4 treatment...like 2, 4, 6, 12, 24 or... hours?

I performed a qPCR for one gene . The results shows 4 samples peak at different Tm in melt curve what could be the reason?

I performed a qPCR for one gene in THP1 cell line cDNA (32 samples and 6 standards). The results shows 3 samples and one standard peak at different Tm in melt curve. What could be the reason?
