Does anyone know how to disrupt a transcript by spacing only in mammalian cells? i.e. in promoter, 5' UTR, 3' UTR, between poly A.

I need the threshold to be 300-600 base pairs and would like to completely shut off transcript at that point. I.e. not gradually reduce efficiency but hit that threshold and disrupt transcript...
