The recommended dilution for the use of antihuman CD3 for t cell activation is 1:1000 but I used it at dilution of 1:100 will that affect the assay?

I used same dilution for negative as well as positive control

In my Fluorospot Assay to detect antigen secreting cells I coated few well with only PBS without antigen. Why I am getting Spots in those wells?

I am coating the well with blank 1X PBS and then Block it with complete RPMI media and after blocking give these well PBS wash and seed the stimulated PBMCs which have Antigen Secreting cells.
