Can anyone tell that for 250kDa protein (NM2) how much time need to transfer and in which volt or Amp ? And how much time I need to run the protein ?

My western blot is not coming properly. I am assuming that due to overnight transfer the protein band is not coming in chemidoc Biorad. Or when coming too much hazy it is. Can anyone please help...
