Could you please help me to find out the suitable way to count and adjust zymosan particles for the phagocytosis assay in macrophages?

I am using zymosan A bioparticles ( molecular probs) Alexa flouor 594 . cat# z- 23374 with the opsonizing reagent cat# z-2850 I would appreciate any comments and suggestions
