I want to conduct a sensitivity test comparison between two methods of detection which; PCR and LAMP. Which one is better?

Usually for sensitivity, researchers will be using cloned DNA. So that we can test the sensitivity based on DNA copies. However, is there any difference if I'm using purified DNA from PCR product?

How can one solve a false positive result for LAMP reaction, exclude contamination?
