Why do some hard ticks who feed on vertebrate hosts engorge yellow and some dark red?

Does it mean that yellow ticks have only taken up plasma? Will such ticks moult successfully? I want to know the mechanism behind this phenomenon. Please see below picture.

Does anybody know how to store Theileria annulata/lestoquardi infected RBCs long term, in order to be used in in vitro infection of vector ticks?

As the blood is going to be imbibed by ticks in an in vitro feeding system, I'm looking for non-toxic methodology (cryoprotectants).
