A pET28a(+) bearing an insert is transformed into E. coli BL21DE3. But the protein is not expressing with conc. of IPTG and different temperature?

I have cloned a mycobacterial soluble fraction gene into pET28a(+) and transformed into E. coli BL21DE3. I tried 0.1, 0.2, 0.5 0.6, 0.8, 1.0 and 1.5 mM IPTG and 16, 20, 25 and 37C temp., but...
