I need estimate the haplotype diversity in mtDNA - COI in nest of Solenopsis invicta, but I need a method more cheap that sequencing, then I prefer restriction enzyme.

Can anyone answer a question about amplification with a probe?

The amplification uses 2 primers plus the probe (with the fluorescent molecule). I would like know: does the probe amplify within the amplicate fragment (by 2 primers)?
