What initial conc. should be used to anneal two 63 bp oligos? I want to do ligation of a ds oligo with a vector, what conc. of oligo should I use?

There is some secondary structure formation when I analysed it in silico, so I am going to go for gradual cooling.

I want to anneal 2 oligos but they are forming a hairpin. I will denature them at 95 degrees, but will the hairpins form again as I lower it to room temp?

Each oligo is 45 mer long.
