For MMP 1, what range of collagen or casein need to be added as substrate in zymograph?

In Collagen zymography gel, the background itself is not stainig by commassie stain? I could not get collagenic bands in casein zymography too? How to overcome these problem?

How can I decide on addition or removal of water molecules during ligand preparation?

I am working on MMPs. Before docking the MMPs with a ligand, How to prepare it? What are the conditions/parameters to be decided for ligand preparation? On what basis those conditions are designed ?
