Should I expect poor CD4 staining of NHP PBMCs following expansion with anti-CD3/anti-CD28 and subsequent stimulation with PMA/Ionomycin?

Seeing CD4 staining levels lower than isotype control by flow cytometry. Cells were previously expanded, stimulated, then fixed and stored at -80C.
