In a infection plant assay, is it really necessary to collect newly emerged leaves from a plant, to prove (by PCR) that it infected? Why?

I work with tomatoes and geminivirus, and I would like to do an infection assay. I heard ios better to collect newly emerged leaves. Why?Thanks
