If a gene is expressed in different types of cells (eg Paneth cells and epithelial), can the gene be mutated in one type of cell and not the other?

Let's say a gene is expressed in two types of cells. Can it be mutated in the first type and be wild-type in the other type of cell?

How do you remove the FLAG-Tag from a "flagged" vector plasmid?
