Can restriction enzymes only be added to the end of a primer or is it possible to add it within the primer sequence?

Testing primers previously designed where restriction enzymes were located in the middle of the primer sequence but not sure whether this is actually possible.

When doing antioxidant assays (plants), would one need to generate a standard curve for the specific assay?

I will be doing the DPPH. SOD, CAT , AP and GR assays and was wondering how I'd determine if there is any effect when comparing my plant samples subjected to different abiotic and biotic stresses.
