Why does the expression of MHC 1 decrease when lymphocytes are stimulated with LPS ?

I have noticed whilst using Beta 2 Microglobulin as a reference gene, the expression reduced significantly whilst doing PCR, can any one explain this? Cells were stimulated with LPS.

In macrophage activation ( RAW264.7) why does the proliferation of macrophages stop when they are activated with LPS?

There is an increase in NO production and increase in phagocytosis (increased uptake of Neutral Red) but Alamar Blue assay shows a drop in cell numbers. Any explanation?

What is the best way to precipitate proteins, from an extract, which also has polysaccharides?

I can´t use TCA as this will damage cells and I will be going on to test this fraction against cells.
