I am trying to polymerise microtubules under TIRF but I am unable to see any dynamicity. What could be the possible reasons for no dynamicity?

The final concentration of the tubulins is 30 uM and GTP is 1 mM

I tried to visualize reconstituted microtubules in IRM. The MTs came together and formed 'flower-like' structures or stayed in bundles. Explanation?

I tried to reconstitute them using GTP, GMPCPP & Taxol separately. Every time I observed the same. It was hard to spot any single MT.
