I am trying to insert a 279 bp fragment into a 8500 bp vector. Many colonies appear after ligation but no p-DNA can be extracted. Any solutions?

I am trying to insert a 279 bp fragment into a 8500 bp vector (pMRS127). After transformation of the ligation product, there appear to be many colonies growing on the antibiotic selection media....
