Cryosection zebrafish larvae WITHOUT fixation?

I need using cryostat with NO fixing embryos. Any suggestion for getting more "robust" sample? I have some tissue problem after sectioning... Thank you so much!

Has anybody a protocol to determine the AChE activity in larvae zebrafish brain?

Even following the instruction of Karnovsky and Roots (1964), I still do not know how long I should fix the larvae and in what fixative. Could anyone help me? Thank you!
