I've transfected HEk293, HEk293T, Cos-7 cells with 2 different plasmids. By PCR I can confirm that the DNA is there but by Western Blot there is no overexpression of the proteins. Any ideas why...

How should long I incubate my cells with an inhibitor before adding the substrate?

I don't see much difference in 2-NBDG uptake with or without phlorizin? I use to incubate HEK293 cells with phlorizin for 10-15 min. Should I leave it longer before adding 2-NBDG?
