What is the best method for quantifying the total protein concentration in a ligament sample?

I am concerned that using a Lowry assay may not capture all protein content in a ligament sample. Any suggestions for a method? I will be using total protein to standardize a Collagen 3 ELISA.

IHC with DAB appears to be a heavily debated topic when it comes to quantification. If not possible to quantify, what inferences can be made instead?

In other words, is there an acceptable quantitative methods to obtain information from DAB visualized antigens? If not, what information can be semi-quantified or inferred from this IHC method?
