Just out of curiosity: Does anybody have an experience of what happens to your protein samples if you overboil them in Laemli buffer (with DTT)?

I had to boil my samples (imported radiolabelled proteins into mitochondria followed by lysing them) prior to loading on an SDS PAGE. Usually I do it for 3 mins at 96°C and it always works well....
