Hi

im trying to calculate mg GAE/g extract value. Im trying to use the following equation:

(mg GAE/g dry extract)=C* V/M

Where C is mg/ml (based on gallic acid calibration curve)

V is ml volume 

and M is mass

I initilay dissolve my dried extract in dionized water then take 40 ul(0.1g/ml) and add to water , FC and Na2Co3 making a total volume of 4 ml.

 my question is V is whether the total amount of reagents used during the assay or only the volume of extract itself (what i mean is that during the preparation my total volume including FC reagent , soudioum carbonate and water is 4 ml but my extract is only 40 microliter.Which one should i take into account
