Diabetes was induced by a single intraperitoneal injection of STZ (60 mg/kg) in 0.1 mol/l citrate buffer (pH 4.5) to the overnight fasted rats. Non-diabetic rats were injected with a similar volume of citrate buffer alone. Two days after the STZ injection, fasting glucose levels were determined in a blood sample taken from the tail vein. Diabetes was confirmed when blood glucose levels were at least 250 mg/dl for 2 consecutive days.
