I did QqQ LC-MS/MS of a single xanthone as per the conditions reported in two research papers. But it was not eluting from the column even after the prescribed time.
What are the best conditions for the effective analysis.
Why not provide us with the DETAILED method analysis condition (HPLC conditions, all of them) you used to retain and try to elute your sample? You should have been able to change the elution conditions to "force" the sample off the column.
Once you discover which conditions DO elute the sample from the column, than you can work back to developing conditions which first retain the sample for an appropriate amount of time and elute it to achieve an acceptable k Prime and resolve it away from any other peaks in your sample ("The process of HPLC Method development").
