i isolated protoplast from pomegranate leaves using 0.4M mannitol containing enzyme solution and washed with W5 and the pellet resuspend in 3 ml of W5 and overlay on 25 percent sucrose everything got sinked without forming protoplast band, the pellet has protoplast
