I am studying mouse NK cells infiltrating MC38 colon cancer or B16 melanoma inoculated into the syngeneic C57B/6 mice. To this end, I need to identify the NK cells that are naive, inhibited, activated, post-activation, memory, exhausted, etc. What are the best (surface) markers for these subsets? Any advice highly appreciated.
As for this quetions, it make me confusion . do you mean that you want to identify the NK subsets for the mouse species? if that, NK1.1 is good for C57 mouse but not all mouse strains . CD335 and CD49b will be good identify excluding the C57 mouse. like Balb/c ，C3H and so on.....
In NK, there are only differences of marker under different species in Mouse.That is, mature and immature NK. There is also divided into ThNK 1/2/3, and NKreg and NKDC. We have no obvious ideas about how to identify the activated and exhausted subsets of mouse NK cells.
We didn't use this on NK, only use this on T cell for the exhausted marker. If you want to identify the activated NK and exhausted NK , you may try to evaluate some cytokines or relevant downstream proteins.
