I'm hoping to use iDisco to clear and stain some mouse DRG. These tissues have already been treated to a sucrose gradient (10/20/30) and are currently in 30% sucrose.
Can I safely start iDisco by placing them in a methanol gradient, and if so, should I begin with 20% methanol or something closer to 100% methanol avoid rapid rehydration?
