In the field of olive oil luteolin content could be checked by rp-hplc analysis. In general there is a previous extraction phase of polyphenols (for example by a mixture of methanol/water) and then the methanolic phase, which contains the polyphenols, is injected into the hplc system. If your detector is not a destructive one (and i think that in general it isn't a destructive one in hplc), you can try to collect luteolin after elution. You could check on web for polyphenols extraction protocols from olives/olive oil as an example.
Thank you Dr Giacomo for the answer. I understand what you say about extraction of luteolin or generally flavonids based Hertog 1993 and Abdel-Moemin 2004 methods. But i need a clear procedure to isolate luteolin and a test to confirm the presence of it. Thank you again for participation
you need to study, physical and chemical properties to extract , at first, flavonoids , then prepare specific column to bind luteolin (prepare solvent system)
I think you must prepare mix solvent to gain luteolin
Try to extract Luteolin from the crushed leaves using methanol-water mixtures (you may use different organic solvent with different polarity), then you can identify and isolate using RP-HPLC.
