You can measure the irradiance (W/m2) at different wavelengths (nm) that will give you the energy of photons. Finally, you can sum up the energy of photons for the entire wavelength range to calculate the photon flux (moles of photon/m2s). This photon flux value is essential to calculate the quantum yield.

Just want to add: Total number of photons (m2/s)=(total irradiance)/(total energy of photons)
