I'm using Fluro-Jade C to stain dead neurons in cryo-sections. As usual, I use a protocol similar to that one described at the merck website: http://www.merckmillipore.com/DE/de/product/Fluoro-Jade-C,MM_NF-AG325
Before placing the slides into 70 % EtOH, slices should be immersed in basic EtOH (1 % NaOH in 80 % EtOH). And I'm wondering what that step is for? Why is it basic EtOH? The FJC dye requires acidic enviroment. If anyone has an explanation, perferably including a reference, I would be happy to hear about it!
