I was able to obtain bactericidal activity using certain antimicrobials in vitro at specific concentrations, now I plan to test them in vivo using a mouse model. Is there a formula (or a method) to help me convert the concentrations I obtained in vitro to a dosage that is appropriate in vivo?
The in vitro concentration that kills bacteria should be achieved in vivo. This means that you have to find out drug absorcion into plasma and its distribution in biological fluids, plasma half life, union to plasma proteins, possibility of its diffusion through the blood brain barrier, etc. In one word, you have to find out distribution, transortation, metabolism and toxicity of the drug.
I will suggest that you should remember to put into consideration the body size and weight of the experimental animal ( mouse) to ease the conversion from in vitro to in vivo.
You go by pharmacodynamics of the agent. Experimental pharmacology books will be helpful for your purpose.
