Yes, HP-20 can be part of a purification procedure. It is a polymeric reverse phase rather than C18, but it should work.

I'd use a gradient with increasing concentrations of methanol in water to 100% methanol.

Diaion HP20 can be used for the purification of flavonoids provided your feed is aqueous. The percentage of polar organic solvent such as methanol, ethanol and isopropanol should not be more than 20% in the feed to favour more binding on the resin. After binding you can elute it with increase in percent methanol or ethanol either in step gradient or linear gradient mode.
