Hi, I am working on phytoremediation of industrial effluent using aquatic plants; So I have to analyse of phytoremediated plant leaf after phytoremediation process to know the surface morphology of leaf. In that, I don't have much of idea about sample fixation for SEM analysis. So kindly suggest me the protocol or procedure.
A few parts of leaves cut randomly were fixed in 4% glutaraldehyde in cacodylate buffer for 24 h, and then transferred to sucrose 0.25 M for 24 h. After this time these pieces are dehydrated up to 100% ethyl alcohol.Increasing series of alcohol from water to 100% ethyl alcohol (10%, 20%, 40%, 60%, 80%, 90%, 100% 100%, with interval of 2 hours, (there are automatic devices that do this dehydration). Complete desiccation was achieved by the CO2 critical point technique. Samples then were covered by a 16 nm thin layer of molecular gold using an sputter coater.
