I don't know what was the dose you administered. I'm sure that you'd measured the dose by doing acute toxicity studies. In case of using a very high toxic dose, where mortality rate is higher, reduction of dose is highly suggested. For orally administration of ethanol you must select lower dose. Even the drug or extract you want to experiment the effect it can be lethal acting as agonist. Make sure why the rats died.
Most of the researchers follows CCl4 and Paracetamol induced hepatotoxicity model in rat. Also, Ethanol is used commonly to induce hepatotoxicity. I think you have to verify the dose and duration of ethanol administration to the rats with literature to prevent the mortality rate.
I have searched the literature and selected a dose of 2ml/kg of 40% alcohol twice daily. I tried to find out the reason of mortality, the histopathological findings suggested that there is hemorrhagic ulcer formation in gastric tissue of animal. What could we do to prevent this or reduce this problem?
